A highly sensitive flexible sensor based on the MXene/αFe2O3 composite structure applied for the simultaneous detection of glucose and uric acid

Sensors based on inorganic nanomaterials for the detection of human body fluids show great potential in real-time detection and treatment of human health due to their advantages of small size and easy to wear. Nevertheless, most of the existing sensors have the disadvantage of detecting relatively simple substances and suffer from low sensitivity, which undermines their practical applicability in body fluids detection. In this study, based on the transition metal carbide (Ti₃C₂T_x, MXene)/αFe₂O₃ grid composite structure, a highly sensitive flexible sensor is proposed, which can realize accurate and simultaneous detection of glucose and uric acid (UA) concentration. The sensor exhibits outstanding durability, enabling it to withstand prolonged immersion in water and exposure to high flow velocities. More importantly, the sensor has extremely high sensitivity for both glucose (2.14 mA mM^-1 cm^-2, 5.5 times the mean value) and UA (2.56 μA μM^-1 cm^-2, 3.5 times the mean value). It also has the advantage of low detection limits (0.27 μM for glucose and 0.31 μM for UA) and a wide linear detection range (1 mM-40 mM for glucose and 10 μM-900 μM for UA) compared to existing sensors for body fluids detection. In addition, the sensor has excellent repeatability, long shelf life (over 40 days) and excellent immunity to interference (accurate detection of glucose and UA in the presence of multiple metal ions as well as organics in human body fluids). This study establishes the groundwork for developing a production-ready and highly sensitive blood detection sensor capable of real-time monitoring.

Role of glutamatergic signaling in the acquisition and expression of learned sugar preferences in C57BL/6 mice

C57BL/6 (B6) mice learn to prefer glucose or sucrose to initially isopreferred or even more preferred nonnutritive sweeteners due to the postoral appetite stimulating (appetition) actions of glucose. Recent evidence indicates that specific duodenal neuropod cells transmit the glucose appetition signal to the brain via glutamatergic synaptic connections with vagal afferents. The present study found that intraperitoneal pretreatment with a glutamatergic receptor antagonist cocktail (kynurenic acid (KA)/D-2-amino-3-phosphonopentanoic acid (AP3)) in B6 mice did not block the expression of their learned preference for 8% glucose solution over an initially-preferred 0.1% sucralose + 0.1% saccharin solution. However, acquisition of the glucose preference was blocked by drug treatment during 1-h training sessions with the two sweeteners. Systemic KA/AP3 injections also did not block the expression of the learned preference for a 10.6% sucrose solution over a 0.6% sucralose solution. Drug effects on the acquisition of the sucrose preference were not determined because sucrose, unlike glucose conditioning, required 24-h training trials. The findings that the 1-h training regimen conditioned 8% glucose, but not 10.6% sucrose, preferences suggest that glucose has more potent appetition actions. This was confirmed by the finding that B6 mice learned to prefer 10.6% glucose to 10.6% sucrose after 1-h or 24-h training despite an initial strong sucrose preference. This action can be explained by 10.6% sucrose's digestion in the gut to glucose and fructose with only glucose activating the gut-brain appetition pathway.

Antimicrobial effect of sulconazole in combination with glucose/trehalose against carbapenem-resistant hypervirulent Klebsiella pneumoniae persisters

The emergence and rapid dissemination of carbapenem-resistant hypervirulent Klebsiella pneumoniae (CR-hvKP) pose a serious threat to public health. Antibiotic treatment failure of K. pneumoniae infections has been largely attributed to acquisition of antibiotic resistance and bacterial biofilm caused by the presence of antibiotic persisters. There is an urgent need for novel antimicrobial agents or therapy strategies to manage infections caused by these notorious pathogens. In this study, we screened a collection of compounds that can dissipate bacterial proton motive force (PMF) and intermediate metabolites that can suppress antibiotic tolerance, and identified an antifungal drug sulconazole which can act in combination with glucose or trehalose to exert strong antibacterial effect against starvation-induced CR-hvKP persisters. Investigation of underlying mechanisms showed that sulconazole alone caused dissipation of transmembrane PMF, and sulconazole used in combination with glucose or trehalose could significantly inhibit the efflux activity, reduce NADH and ATP levels, and cause intracellular accumulation of reactive oxygen species (ROS) in CR-hvKP persisters, eventually resulting in bacterial cell death. These findings suggest that the sulconazole and glucose/trehalose combination is highly effective in eradicating multidrug-resistant and hypervirulent K. pneumoniae persisters, and may be used in development of a feasible strategy for treatment of chronic and recurrent K. pneumoniae infections.

销售额超40亿的化药品种，你绝对想不到
