编辑丨王多鱼 排版丨水成文 利用合成致死（Synthetic Lethality）特异性杀伤肿瘤细胞，是近年来肿瘤临床治疗的一项重要尝试。特别是靶向 BRCA1/BRCA2 突变的 PARP 抑制剂（PARPi），自问世以来便展现出显著的临床疗效，为基于合成致死原理的靶向药物开发提供了积极的实践验证。 然而，PARPi 的应用有其局限性，即便在 BRCA1/2 突变的肿瘤细胞中，也并非所有病例均能对其产生敏感应答。究其原因，除了耐药机制的存在，还在于 BRCA1/2 所参与的生物学过程比以前学术界所知的更为复杂。近年来，BRCA1/2 被发现在复制叉稳定性维持中发挥关键作用，然而细胞进行复制叉稳定性维持的分子机制还未得到清晰揭示。 2025年1月14日18时，浙江大学刘婷教授团队、方东教授团队及黄俊教授团队（博士生谢海华、宋礼志为共同第一作者）合作，在 Nature 子刊 Nature Chemical Biology 上发表了题为：Synergistic protection of nascent DNA at stalled forks by MSANTD4 and BRCA1/2-RAD51 的研究论文。该研究揭示了一条与 BRCA1/2 合成致死的新通路。 在这项最新研究中，研究团队发现了一个具有与端粒结合蛋白 TRF1/TRF2 类似的 Myb/SANT-like DNA 结合结构域的新蛋白——MSANTD4（Myb/SANT-like DNA-binding domain-containing protein 4），能结合于停滞复制叉翻转臂的 3'-ssDNA 末端，特异性阻碍 RPA-WRN/BLM-DNA2 复合物对翻转臂上新生 DNA 的过度切割，从而维持停滞复制叉的稳定性。该通路完全独立于传统已知的由 BRCA1/2-RAD51 介导的停滞复制叉保护机制，因此与 BRCA1/2 合成致死。 该研究首次报道了 MSANTD4 在复制叉稳定性维持中的功能及分子机制，为深入理解复制叉稳定性维持的分子机制以及肿瘤治疗提供了全新视角。 在高等真核生物中，复制叉翻转（Fork reversal） 是细胞应对复制压力的一种进化上高度保守的机制。在此过程中，“三叉”结构的复制叉在转位酶（例如 SMARCAL1、ZRANB3、HLTF 和 PICH），以及 DNA 拓扑异构酶 TOP2A 和 SUMO E3 连接酶 ZATT 等因子的协作下，被重塑为更稳定的“四叉”结构，从而维持复制叉的动态平衡。然而，复制叉翻转臂末端由于其结构类似于单一末端的 DNA 双链断裂（One-ended DSB），极易受到核酸酶攻击，从而引发新生DNA链的降解，导致基因组不稳定性。 已有研究表明，BRCA1/2 介导的 RAD51-DNA 核纤丝（RAD51 filament） 能够保护停滞复制叉的翻转臂，起到维持复制叉稳定性的作用。然而，现有证据显示，BRCA1/2-RAD51 轴并不能完全抵御多种核酸酶的攻击，这表明可能存在独立于 BRCA1/2-RAD51 的其它保护机制，有待进一步研究和揭示。 在解旋酶 WRN/BLM 等的协助下，核酸酶 DNA2 可修剪翻转复制叉的末端，产生带有  3'-单链DNA突出末端（3'-overhanged DNA）的结构。这一结构是复制压力缓解后，复制叉通过同源重组（Homologous recombination，HR）实现重启的关键。然而，若这一切割过程失控，将导致复制叉的降解，并显著增加基因组不稳定性的风险。因此，如何精确调控这些复合物的活性，成为实现停滞复制叉精准调控和有效保护的关键科学问题。 研究团队发现，MSANTD4 蛋白在复制压力下能够被招募至停滞复制叉，并特异性结合 3'-overhanged DNA。该蛋白通过拮抗 BLM 或 WRN 对复制叉翻转臂双链 DNA 的解旋活性，有效阻止了 DNA2 对新生 DNA 的降解。与这一作用一致，MSANTD4 的缺失会导致细胞内新生 DNA 降解显著增加，同时提升细胞对复制压力的敏感性。此外，由于 MSANTD4 保护翻转复制叉末端的机制与传统的 BRCA1/2-RAD51 轴完全不同，研究者还发现，MSANTD4 的缺陷能够进一步加剧 BRCA 缺陷细胞中新生 DNA 的降解现象，并显著提高这些细胞对复制压力的敏感性，展现出“合成致死”的效应。 综上所述，该研究首次报道了 MSANTD4 在复制叉稳定性调控中的关键功能与作用机制；系统阐明了 MSANTD4 保护停滞复制叉新生 DNA 链的分子生物学过程；同时揭示了一种 MSANTD4 与 BRCA1/2-RAD51 协作的新型复制叉保护模式。该协同机制能够有效防止核酸酶对停滞复制叉翻转臂中新生 DNA 的过度降解，为 BRCA1/2 突变相关肿瘤的“合成致死”疗法开发提供了潜在的全新靶点。 论文链接： https://www-nature-com.libproxy1.nus.edu.sg/articles/s41589-024-01833-9 设置星标，不错过精彩推文 开放转载 欢迎转发到朋友圈和微信群  微信加群  为促进前沿研究的传播和交流，我们组建了多个专业交流群，长按下方二维码，即可添加小编微信进群，由于申请人数较多，添加微信时请备注：学校/专业/姓名，如果是PI/教授，还请注明。 点在看，传递你的品味

Root hairs represent a low-cost strategy to enhance nutrient uptake because they can significantly increase the nutrient-acquiring surface of plant roots. While primary and lateral roots are stimulated to elongate when plants grow under mild nitrogen deficiency, the existence of such a foraging response for root hairs and its underlying regulatory mechanism remain elusive. Now, researchers have revealed a framework composed of specific molecular players meditating auxin synthesis, transport and signaling that triggers root hair elongation for nitrogen acquisition. Root hairs represent a low-cost strategy to enhance nutrient uptake because they can significantly increase the nutrient-acquiring surface of plant roots. While primary and lateral roots are stimulated to elongate when plants grow under mild nitrogen deficiency, the existence of such a foraging response for root hairs and its underlying regulatory mechanism remain elusive. Now, an international research team led by IPK Leibniz Institute revealed a framework composed of specific molecular players meditating auxin synthesis, transport and signaling that triggers root hair elongation for nitrogen acquisition. The results have been published in the journal Current Biology. Plants have evolved diverse adaptive strategies to optimize the exploration of light and soil resources from their environments. One of the most prominent adaptive responses is the developmental plasticity of their root systems, which enables plants to efficiently forage nutrient pools that fluctuate in space and time. As a crucial component determining the active root surface, root hairs represent a powerful morphological trait to improve water and nutrient acquisition. Root hairs are extensions of epidermal cells that effectively enlarge the root surface area and facilitate soil exploration for water and nutrients. Root hair development starts with cell fate specification, which determines whether an epidermal cell becomes a hair-forming cell (trichoblast) or a hairless cell (atrichoblast). "In our study, we first showed that nitrogen deficiency strongly stimulates root hair elongation in the model species Arabidopsis thaliana and that this response is crucial for plant fitness under low nitrogen," explains Prof. Dr. Nicolaus von Wirén, head of the department "Physiology and Cell Biology" at IPK Leibniz Institute. "And we then integrated transcriptomics, molecular genetics and cell biology approaches to establish that low nitrogen-induced root hair elongation builds on a spatially coordinated auxin signaling cascade that enters the root hair developmental program via the epidermal transcriptional module RHD6-LRL3." The signaling cascade consists of three stages. Through upregulation of the enzymes TAA1 and YUCCA8, low nitrogen increases auxin accumulation in the root apex (step 1, synthesis). Auxin is then directed shootward to the root hair differentiation zone via the transporters AUX1 and PIN2 (step 2, transport). Upon arrival, auxin activates the transcription factors ARF6 and ARF8 to promote the transcriptional module RHD6-LRL3, which is confined to the epidermis and steers root hair elongation in response to low nitrogen (step 3, signaling). "Overall, our results suggest that root hair elongation represents an additional systemically-induced foraging response strategy to explore the soil for nitrogen, gaining even more relevance as nitrogen deficiency becomes more severe," says Dr. Zhongtao Jia, first author of the study. "These findings add a new feature to our understanding of root foraging responses in low-nutrient environments and expand the mechanistic framework of hormone-regulated nutrient sensing in plant roots." Since more abundant and longer root hairs represent a low-cost strategy for plants to bring roots in contact with soil nutrients, understanding how root hair development is regulated by nitrogen provides new breeding targets for developing crops with enhanced nitrogen-uptake efficiency.
