Deubiquitinase that mediates deubiquitination of target proteins such as BECN1, MITF, SKP2 and USP10 and is involved in various processes such as autophagy, endoplasmic reticulum-associated degradation (ERAD), cell cycle progression or DNA damage response (PubMed:21571647, PubMed:32772043, PubMed:33592542). Component of a regulatory loop that controls autophagy and p53/TP53 levels: mediates deubiquitination of BECN1, a key regulator of autophagy, leading to stabilize the PIK3C3/VPS34-containing complexes. Alternatively, forms with NEDD4 a deubiquitination complex, which subsequently stabilizes VPS34 to promote autophagy (PubMed:32101753). Also deubiquitinates USP10, an essential regulator of p53/TP53 stability. In turn, PIK3C3/VPS34-containing complexes regulate USP13 stability, suggesting the existence of a regulatory system by which PIK3C3/VPS34-containing complexes regulate p53/TP53 protein levels via USP10 and USP13. Recruited by nuclear UFD1 and mediates deubiquitination of SKP2, thereby regulating endoplasmic reticulum-associated degradation (ERAD). Also regulates ERAD through the deubiquitination of UBL4A a component of the BAG6/BAT3 complex. Mediates stabilization of SIAH2 independently of deubiquitinase activity: binds ubiquitinated SIAH2 and acts by impairing SIAH2 autoubiquitination. Regulates the cell cycle progression by stabilizing cell cycle proteins such as SKP2 and AURKB (PubMed:32772043). In addition, plays an important role in maintaining genomic stability and in DNA replication checkpoint activation via regulation of RAP80 and TOPBP1 (PubMed:33592542). Deubiquitinates the multifunctional protein HMGB1 and subsequently drives its nucleocytoplasmic localization and its secretion (PubMed:36585612). Positively regulates type I and type II interferon signalings by deubiquitinating STAT1 but negatively regulates antiviral response by deubiquitinating STING1 (PubMed:23940278, PubMed:28534493).

Rhythmic TDP-43 affects RNA splicing of USP13, resulting in alteration of BMAL1 ubiquitination

Circadian rhythm disorders are common characteristics of neurodegenerative diseases. The pathological aggregation of transactive response DNA-binding protein 43 (TDP-43) is associated with multiple neurodegenerative diseases, such as amyotrophic lateral sclerosis. However, the relationship between TDP-43 and circadian rhythm remains unknown. Here, we found that TDP-43 is rhythmically expressed both in vivo and in vitro. TDP-43 knockdown affected the expression of circadian genes, including BMAL1, CLOCK, CRY1, and PER2, and impaired autonomous circadian wheel behavior, cognitive functions, and balance abilities in mice. Furthermore, TDP-43 knockdown induced aberrant splicing of ubiquitin-specific peptidase 13 (USP13) and blocked USP13 rhythmic expression, enhancing the ubiquitination of BMAL1. Meanwhile, TDP-43 knockdown altered the rhythmic expression of phospho-AMPKα (Thr172) and platelet-type phosphofructokinase (PFKP), which may change cellular glucose uptake and ATP production. Our findings further the understanding of the role of TDP-43 dysfunction in circadian rhythm disruption in neurodegenerative diseases and provide new mechanistic evidence supporting the interaction between circadian rhythm disruption and neurodegeneration.

Ubiquitin specific peptidase 13 protects against inflammation-associated joint injury in collagen-induced rheumatoid arthritis mice by targeting TRAF6

Rheumatoid arthritis (RA) is a chronic inflammatory autoimmune disease with complex pathogenesis. Ubiquitin-specific protease 13 (USP13), a member of the deubiquitinating enzyme (DUB) superfamily, plays diverse roles in cellular events. This study investigates the role of USP13 in RA, revealing its significant downregulation in peripheral blood mononuclear cells (PBMCs) from RA patients compared to healthy individuals. USP13 expression negatively correlates with RA characteristics, including erythrocyte sedimentation rate (ESR), C-reactive protein (CRP), and rheumatoid factor (RF). Consistent with these findings, USP13 levels were reduced in knee joint synovial tissues of collagen-induced arthritis (CIA) mice and lipopolysaccharide (LPS)-stimulated bone marrow-derived macrophages (BMDMs). Mechanistically, USP13 directly interacts with tumor necrosis factor receptor-associated factor 6 (TRAF6), reducing its K63-linked polyubiquitination and thereby inhibiting TRAF6 expression. This interaction restrains nuclear factor κB (NF-κB) signaling, leading to a marked attenuation of LPS-induced inflammatory responses in BMDMs. Importantly, the anti-inflammatory effects of USP13 over-expression are largely dependent on TRAF6 suppression. In vitro, USP13 over-expression in BMDMs inhibits the proliferation of fibroblast-like synoviocytes (FLS) and osteoclastogenesis. In addition, USP13-overexpressing BMDMs in CIA mice significantly alleviates RA development, as evidenced by reduced synovial hyperplasia, inflammatory cell infiltration, cartilage destruction, and bone loss. These findings highlight the essential role of USP13 in macrophages during RA progression and reveal its therapeutic potential by targeting TRAF6 signaling.

Ubiquitin-specific protease 13 regulates FcεRI-mediated mast cell activation and allergic inflammation via SYK protein modulation

Mast cells (MCs) are therapeutic targets for high-affinity IgE Fc receptors (FcεRI)-mediated allergic responses. Deubiquitinating enzymes (DUBs), including ubiquitin-specific protease 13 (USP13), are involved in multiple inflammatory processes. This study aims to reveal USP13's role in FcεRI-mediated MC activation and its underlying mechanisms. Our results showed USP10/13 inhibitor spautin-1 inhibited IgE-mediated MC activation, as evidenced by a reduction in the release of β-hexosaminidase (β-hex) and histamine and decreased expression and secretion of inflammatory cytokines. Spautin-1 also attenuated inflammatory processes in IgE-mediated passive cutaneous anaphylaxis (PCA) and ovalbumin (OVA)-induced active systemic anaphylaxis (ASA) models. Furthermore, knockdown of USP13 by short hairpin (sh)RNA diminished IgE-induced MC activation. Protein-protein interactions assays showed that USP13 interacted with the co-immunoprecipitated protein spleen tyrosine kinase (SYK) and deubiquitinated SYK. USP13 bound the kinase domain of SYK and removed its K63-linked polyubiquitination chain, yielding a more stable SYK protein. Importantly, 2-methoxyestradiol (2-Meth) was identified as a potential inhibitor of USP13 and inhibited FcεRI-mediated MC activation effectively in vitro and in vivo. In conclusion, it elucidated the molecular mechanism by which USP13 regulated SYK stability in MCs. The USP13-SYK axis may serve as a therapeutic target for treating FcεRI-mediated activation of MCs and associated inflammatory responses.

引言胶质母细胞瘤是最致命的恶性肿瘤之一，表现出明显的肿瘤内异质性，其中胶质母细胞瘤干细胞（GSCs）对肿瘤发生和治疗抵抗起着重要作用。泛素连接酶和去泛素化酶的药物调节剂正在开发用于癌症和其他疾病。2024年3月19日，西湖大学谢琦，深圳湾实验室李磊和匹兹堡大学Jeremy N. 性别是除年龄和APOE ε4以外的散发性AD的重要风险因素。本研究结果提示，女性大脑中USP11的生理性表达高于男性，更容易受tau蛋白负担的影响。若能抑制USP11介导的tau蛋白去泛素化可能会为A 阿尔茨海默病（AD）是一种神经退行性疾病，随着人口老龄化，AD患者人数越来越多，现有研究显示，女性患AD的风险约为男性的两倍[1]。造成此性别差异的因素一直存在争议。有观点认为，可能是因为有更多的女性活到老年，使得女性患者绝对数自然大于男性[2]。但是，也有研究指出女性特有的激素变化是其具有AD高风险的原因之一[3]。截至目前，女性高易感AD的分子机制尚未得到阐明。最近，美国凯斯西储大学医学院Jung-A.A.Woo和David E.Kang研究团队在著名期刊Cell上发表重要研究成果[4]。该研究提出了X染色体基因表达的泛素特异性肽酶11（USP11）可能是导致女性AD风险升高的原因。USP11可以使tau蛋白去泛素化，从而导致了tau蛋白的病理性聚集。更重要的是，研究人员发现雌性小鼠和女性大脑中有着更高的USP11水平，并进一步验证了敲除usp11可以保护雌性小鼠免于tau蛋白病理影响，从而改善认知障碍。若能抑制USP11介导的tau蛋白去泛素化，会减少女性对tau蛋白病理的易感性。这可能为降低女性患AD的风险提供了一个方向。论文首页截图让我们看一下这项研究是如何展开的。 tau蛋白的聚集和清除受到各种翻译后修饰的控制，包括磷酸化、甲基化、乙酰化和泛素化[5-8]。其中泛素化主要通过泛素-蛋白酶体系统和自噬-溶酶体通路来完成tau蛋白的清除[9]。一旦tau蛋白的清除过程遭到破坏，那么它就会在神经元内发生病理性聚集，进而导致神经元的破坏。事实上，人类基因组编码大约100种去泛素酶（DUBs），其中近一半是泛素特异性肽酶（USPs）[10]。为了研究USPs在tau蛋白调控中的作用，研究人员进行了功能性小干扰RNA （siRNA）筛选，确定了tau 蛋白的两个阳性调控因子USP13和USP11。鉴于USP11位于X染色体上，所以研究人员提出假设，USP11可能是女性对tau蛋白病敏感的原因之一。研究人员通过细胞转染技术发现， USP11的过表达显著增加了不溶性tau蛋白（神经元纤维缠结的主要成分）而未改变可溶性tau蛋白。这表明USP11促进了tau蛋白的不溶性和病理聚集。 A-B：USP11促进tau蛋白的不溶性 C：USP11增加tau蛋白的病理聚集当研究人员将USP11加到tau蛋白泛素结合物中24小时后，除赖氨酸254外，几乎所有的泛素结合物都从tau蛋白中去除。这也证实了USP11可以去除tau蛋白泛素结合物导致去泛素化。 USP11去除tau蛋白泛素结合物在胚胎发育早期，女性有一条X染色体具有转录活性而另一条将失活。然而，有意思的是，有研究曾指出USP11可以逃避这种失活[11-12]。这也就意味着，相对于男性而言，USP11会在女性中存在过表达的现象。事实也确实如此，研究人员发现，AD女性患者大脑中USP11含量与tau蛋白病理的正相关性确实比男性更高一些( r2=0.5759，P<0.0001)，在多元线性回归分析后，此结果仍有意义。这说明USP11的累积可能是女性tau蛋白病理的驱动因素。 E-F：USP11与女性tau蛋白病理强相关接下来，研究人员在小鼠中敲除usp11来验证对tau蛋白病理的清除影响。结果发现，与未敲除usp11的雌鼠（tauP301S）相比，敲除usp11的雌鼠（tauP301S;usp11-/-）大脑中，不溶性的总tau蛋白和磷酸化tau蛋白减少了50%左右。虽然tauP301S;usp11-/-雄鼠的不溶性tau蛋白也有所减少，但其清除效应不及雌鼠。研究人员进一步对5个月大的雌鼠脑切片进行了电生理记录，来验证USP11的敲除对突触间连接强度的调节影响。研究人员通过双脉冲易化（PPF）和长时程增强（LTP）实验发现，tauP301S雌鼠的场兴奋性突触后电位（fEPSP）比值显著降低，且tauP301S雌鼠脑切片在1小时内LTP严重钝化。然而，tauP301S;usp11-/-雌鼠表现出的fEPSP比值与LTP的诱导和维持是正常的。这表明usp11的敲除可恢复突触可塑性，从而改善认知障碍。 C：5月龄雌鼠PPF的定量分析 D：5月龄雌鼠LTP的定量分析最后，研究人员通过水迷宫的获得性训练和探查试验发现，与tauP301S雌鼠相比，tauP301S;usp11-/-雌鼠发现隐藏平台的时间明显更短，进入靶区平台的时间也更短，空间记忆能力增强，这说明usp11的敲除可以改善雌鼠的空间记忆障碍。然而，usp11的敲除对雄鼠的空间记忆改善意义不大。总的来说，性别是除年龄和APOE ε4以外的散发性AD的重要风险因素。本研究结果提示，女性大脑中USP11的生理性表达高于男性，更容易受tau蛋白负担的影响。若能抑制USP11介导的tau蛋白去泛素化可能会为AD女性患者的治疗提供一个新的方向。参考文献： [1] Rajan KB, Weuve J, Barnes LL, McAninch EA, Wilson RS, Evans DA.
