An HPLC method was established for the determination of Murraya exotica L. leaves flavonoids (JK) in plasma, and the pharmacokinetics of JK as well as the hydroxypropyl-β-cyclodextrin inclusion complex (JK-HP-β-CD) in rats was investigated.Rats received 200 mg·kg-1 of JK and JK-HP-β-CD by oral administration.Blood samples were collected at different times.The concentrations of 5,7,3',4'-tetramethoxyflavone (JA) and 5,7,3',4',5'-pentamethoxyflavone (JB) in plasma were determined, which were the major components of JK.The DAS2.1.1 software was used to calculate the pharmacokinetic parameters.The concentration-time relationships of JK and JK-HP-B-CD were perfectly fitted to two-compartment model.The bioavailability of JA in the inclusion complex was 5.30 times as that in the free drug.The Cmax of JA in inclusion complex was 3.42 times as that in the free drug.The bioavailability of JB in inclusion complex was 4.96 times as that in the free drug.The Cmax of JB in inclusion complex was 2.93 times as that in the free drug.Compared with free drug, the bioavailability of inclusion complex was significantly improved.
