The separation and purification technol. of vancomycin from the fermentation liquor was studied with cation exchange resins in combination with macroporous adsorptive resins.Several types of cation exchange resins and macroporous adsorptive resins were compared on the parameters of load carrying capacity, recovery rate and purification fold to select the optimum resins.The purification technol. for vancomycin was identified as follows: the fermentation liquor of vancomycin was exchanged onto PK206 resin column, next vancomycin was eluted with 0.1 N NH4OH aqueous solution and the fractions 4 and 5 of the eluant (the volume of a fraction was half of the volume of the resin) were collected and purified by CM650 resin column with 0.3 N NH4Cl aqueous solution as the eluent, and then the fractions 6-10 of the eluant from CM650 column were collected, desalted and concentrated by nanofiltration and crystallization to give a final product of vancomycin with a purity of 93.44%.The total recovery of vancomycin was over 64.73%.
