Novel sampling and gas-phase derivatization strategy: Proof-of-concept by profiling ionic polar metabolites using gas chromatography-mass spectrometry

Metabolomic research involves the comprehensive analysis of metabolites in biological samples and has many applications. Gas chromatography-mass spectrometry (GC-MS) is an established and widely used approach for metabolic profiling. However, sample preparation and metabolite derivatization are time-consuming, and derivatization options are limited. We propose gas-solid phase derivatization (GSPD) as a novel sampling and derivatization method that uses a silica monolith substrate and gaseous derivatization reagents for metabolomics using GC-MS. We developed a method to measure the organic acids and sugar phosphates responsible for glycolysis and the tricarboxylic acid (TCA) cycle. GSPD simplifies the sample preparation and can be applied to derivatization reactions that are difficult to perform in solution owing to solvent limitations. The developed method was applied to human plasma and tomato pulp and was shown to have a higher detection performance than the conventional method. This study provides a strategy to simplify sample preparation and expand derivatization options for GC-MS-based metabolomics.

Advantages of online supercritical fluid extraction and chromatography hyphenated to mass spectrometry to analyse plastic additives in laboratory gloves

Plastic additives are introduced in plastic material formulations, along with organic polymers, to offer different properties such as stability, plasticity or color. However, plastic additives may migrate from the plastic material to the content (in case of plastic containers) or to the material in contact with the plastic, like human skin. In the case of plastic medical devices, this migration is of particular interest, as plastic additives may be deleterious to health. In the present paper, we examined the interest of combining supercritical fluid extraction (SFE) to supercritical fluid chromatography (SFC) hyphenated to mass spectrometry (MS) in an online system to characterize plastic additives in laboratory gloves, taken as samples of medical devices. A set of target compounds comprising 18 plasticizers, 4 antioxidants and 2 lubricants was defined and their detectability with MS was examined, where it appeared that electrospray ionization (ESI) provided better detectability than atmospheric pressure chemical ionization (APCI). After examining possible stationary phases with the help of Derringer desirability function, an isocratic chromatographic method (CO₂:methanol 95:5) was developed on Shim-pack UC Phenyl column. The extraction method was examined with a 3-level full factorial design of experiments to optimize the extraction temperature (40 °C) and pressure (200 bar). The online SFE-SFC-MS method was compared to offline methods where the samples were extracted with liquid solvents at atmospheric pressure or high pressure then analysed with SFC-MS. In all cases, offline methods showed significant contaminants (like the oleamide lubricant) issuing from laboratory plastic materials as nitrogen drying station, syringes and filters, while the online method allowed a complete elimination of laboratory contaminations. Furthermore, the online method saved time, solvents and laboratory consumables. It will also show that transferring a compressible fluid from a loading loop is favourable to high efficiency, as the resulting chromatographic peaks are much thinner than when transferring a liquid. Compared to injecting liquid heptane, the efficiency increase was 3.4-fold, while compared to injecting liquid methanol (a common practice in SFC), the efficiency increase was 13-fold. Finally, the additive composition of different laboratory gloves was compared.

Homogeneous B0 coil design method for open-access ultra-low field magnetic resonance imaging: A simulation study

A multimodal brain function measurement system integrating functional magnetic resonance imaging (fMRI) and magnetoencephalography (MEG) is expected to be a tool that will provide new insights into neuroscience. To integrate fMRI and MEG, an ultra-low-field MRI (ULF-MRI) scanner that can generate a static magnetic field (B0) with an electromagnetic coil and turn off the B0 during MEG measurements is desirable. While electromagnetic B0 coil has the above advantages, it also has a trade-off between size and the broadness of the magnetic field homogeneity. In this study, we proposed a method for designing a B0 multi-stage circular coil arrangement that determines the number of coils required to maximize magnetic field homogeneity and minimize the total wiring length of the coils. The optimized multi-stage coil arrangement had an external shape of 600 mm in diameter and a maximum height of 600 mm, with an aperture of 600 mm in diameter and 300 mm in height. The magnetic field homogeneity was <100 ppm over a 210 mm diameter spherical volume (DSV). Compared to a previous two coil pairs arrangement with the same magnetic field homogeneity, the diameter was 1/1.9 times smaller, indicating that the newly designed B0 coil arrangement realized a smaller size and wider magnetic field homogeneity.

Shimadzu Corporation establishes three R&D centers in the US in anticipation of business expansion for pharmaceutical companies

Japan’s Shimadzu Corporation announced this month that its US subsidiary, Shimadzu Scientific Instruments (SSI), has established a new R&D center in the US, and is planning to strengthen its product development capabilities and technology for its work with pharmaceutical companies. The R&D center consists of the Maryland headquarters attached to the SSI headquarters, and new locations near Boston and San Francisco that will work on prototypes developed with the company’s customers. The company’s Maryland headquarters will include a collaboration lab to conduct joint research with customers. Shimadzu Corporation says it plans to increase parent company Shimadzu Group’s sales in North America from 51 billion yen ($330 million) in 2022 to over 70 billion yen ($452 million) in 2025 by leveraging SSI’s bases to expand its business to pharmaceutical companies. “We will work closely with pharmaceutical companies to assess their needs and proceed with the development of products (such as analytical and measuring instruments). In addition to the small molecule drugs that SSI has previously collaborated with pharmaceutical companies on, we will also contribute to the development of biopharmaceuticals,” said a Shimadzu spokesperson.
