Palmitoyltransferase that catalyzes the addition of palmitate onto various protein substrates and is involved in a variety of cellular processes (PubMed:18508921, PubMed:18296695, PubMed:19144824, PubMed:21343290, PubMed:22034844, PubMed:23793055). Has no stringent fatty acid selectivity and in addition to palmitate can also transfer onto target proteins myristate from tetradecanoyl-CoA and stearate from octadecanoyl-CoA (By similarity). In the nervous system, plays a role in long term synaptic potentiation by palmitoylating AKAP5 through which it regulates protein trafficking from the dendritic recycling endosomes to the plasma membrane and controls both structural and functional plasticity at excitatory synapses (By similarity). In dendrites, mediates the palmitoylation of DLG4 when synaptic activity decreases and induces synaptic clustering of DLG4 and associated AMPA-type glutamate receptors (By similarity). Also mediates the de novo and turnover palmitoylation of RGS7BP, a shuttle for Gi/o-specific GTPase-activating proteins/GAPs, promoting its localization to the plasma membrane in response to the activation of G protein-coupled receptors. Through the localization of these GTPase-activating proteins/GAPs, it also probably plays a role in G protein-coupled receptors signaling in neurons (By similarity). Also probably plays a role in cell adhesion by palmitoylating CD9 and CD151 to regulate their expression and function (PubMed:18508921). Palmitoylates the endoplasmic reticulum protein CKAP4 and regulates its localization to the plasma membrane (PubMed:18296695, PubMed:19144824). Could also palmitoylate LCK and regulate its localization to the plasma membrane (PubMed:22034844). The significant regulatory role of palmitoylation modification in cancer-related targets has been demonstrated previously. However, the biological functions of Nrf2 in stomach cancer and whether the presence of Nrf2 palmitoylation affects gastric cancer (GC) progression and its treatment have not been reported. Several public datasets were used to look into the possible link between the amount of palmitoylated Nrf2 and the progression and its outcome of GC in patients. The palmitoylated Nrf2 levels in tumoral and peritumoral tissues from GC patients were also evaluated. Both loss-of-function and gain-of-function via transgenic experiments were performed to study the effects of palmitoylated Nrf2 on carcinogenesis and the pharmacological function of 2-bromopalmitate (2-BP) on the suppression of GC progression in vitro and in vitro. We discovered that Nrf2 was palmitoylated in the cytoplasmic domain, and this lipid posttranslational modification causes Nrf2 stabilization by inhibiting ubiquitination, delaying Nrf2 destruction via the proteasome and boosting nuclear translocation. Importantly, we also identify palmitoyltransferase zinc finger DHHC-type palmitoyltransferase 2 (DHHC2) as the primary acetyltransferase required for the palmitoylated Nrf2 and indicate that the suppression of Nrf2 palmitoylation via 2-bromopalmitate (2-BP), or the knockdown of DHHC2, promotes anti-cancer immunity in vitro and in mice model-bearing xenografts. Of note, based on the antineoplastic mechanism of 2-BP, a novel anti-tumor drug delivery system ground 2-BP and oxaliplatin (OXA) dual-loading gold nanorods (GNRs) with tumor cell membrane coating biomimetic nanoparticles (CM@GNRs-BO) was established. In situ photothermal therapy is done using near-infrared (NIR) laser irradiation to help release high-temperature-triggered drugs from the CM@GNRs-BO reservoir when needed. This is done to achieve photothermal/chemical synergistic therapy. Our findings show the influence and linkage of palmitoylated Nrf2 with tumoral and peritumoral tissues in GC patients, the underlying mechanism of palmitoylated Nrf2 in GC progression, and novel possible techniques for addressing Nrf2-associated immune evasion in cancer growth. Furthermore, the bionic nanomedicine developed by us has the characteristics of dual drugs delivery, homologous tumor targeting, and photothermal and chemical synergistic therapy, and is expected to become a potential platform for cancer treatment.

Constitutive Plasma Membrane Turnover in T-REx293 cells via Ordered Membrane Domain Endocytosis under Mitochondrial Control.

Clathrin/dynamin-independent endocytosis of ordered plasma membrane domains ( o rdered m embrane d omain e ndocytosis, OMDE) can be far more powerful than conventional endocytosis. Nevertheless, it remains controversial how OMDE takes place, in which cell processes it is important, and how it is regulated. Using patch clamp, we have described that OMDE becomes massive in multiple cell types in response to Ca elevations, G protein activation by non-hydrolyzable GTP analogs, and enhanced oxidative metabolism. We describe here that cytoplasmic succinate and pyruvate, but not β-hydroxybutyrate, can induce OMDE of 75% of the plasma membrane in murine marrow macrophages (BMMs) within 2 min. OMDE is decreased by 70% in BMMs lacking the acyltransferase, DHHC5, by treatment with carnitine to shift long- chain acyl groups from cytoplasmic to mitochondrial acyl-CoA's, by bromopalmitate/albumin complexes to block DHHCs, and by the cyclosporin, NIM811, to block permeability transition pores that may release mitochondrial coenzyme A into the cytoplasm. Using T-REx293 cells, plasma membrane loss amounts to 40% with pyruvate or GTPγS, whereby OMDE is then highly sensitive to actin cytoskeleton disruption. Using fluorescent 3kD dextrans to monitor constitutive endocytosis in unperturbed cells without serum or bicarbonate, multiple results suggest that OMDE is constitutively active in T-REx293 cells, but not in BMMs. In T-REx293 cells, substrates that generate metabolites activated by coenyzme A (CoA), e.g. pyruvate and hydroxybutyrate, inhibit endocytosis as expected for a shift of long-chain acyl-CoA to other CoA metabolites. In the presence of bicarbonate, however, pyruvate strongly enhances OMDE, and OMDE is then blocked by β-hydroxybutyrate, bromopalmitate/albumin complexes, and cyclosporines. After pyruvate responses, T-REx293 cells grow normally without evidence of apoptosis. Low concentrations of fatty acid (FA)-free albumin inhibit basal endocytosis in T-REx293 cells, but not BMMs, as do cyclosporines, carnitine, and RhoA blockade. However, constitutive endocytosis (i.e. in the absence of substrates and bicarbonate) is not inhibited by siRNA knockdown of the acyltransferases, DHHC5 or DHHC2, which support OMDE responses activated in patch clamp experiments. We verify biochemically that small CoA metabolites cause a decrease of long-chain acyl-CoA's and that several palmitoylated membrane proteins are depalmitoylated during treatments that decrease long-chain acyl-CoA's. STED microscopy reveals that vesicles formed in constitutive OMDE in T-REX293 cells have diameters of 90 to 130 nm. In summary, palmitoylation-dependent OMDE is likely a major G-protein-dependent endocytic mechanism that can be constitutively active, albeit not in BMMs. It depends on the activities of different DHHC acyltransferases in different cellular circumstances and can be limited by local supplies of fatty acids, CoA, and long-chain acyl-CoA's.

Genetic correlation, shared loci, but no causality between bipolar disorder and inflammatory bowel disease: A genome-wide pleiotropic analysis.

BACKGROUND AND AIMS:

The comorbidity between bipolar disorder (BD) and inflammatory bowel disease (IBD) has been widely reported in observational studies. However, unclear whether this comorbidity reflects a shared genetic architecture.

METHODS:

Leveraging large-scale genome-wide association study (GWAS) summary statistics of BD, IBD and its subtypes, ulcerative colitis (UC) and Crohn's disease (CD), we performed a genome-wide pleiotropic analysis to estimate heritability and genetic correlation, identify pleiotropy loci/genes, and explore the shared biological pathway. Mendelian randomization (MR) studies were subsequently employed to infer whether the potential causal relationship is present.

RESULTS:

We found a positive significant genetic correlation between BD and IBD (r_g = 0.10, P = 7.00 × 10^-4), UC (r_g = 0.09, P = 2.90 × 10^-3), CD (r_g = 0.08, P = 6.10 × 10^-3). In cross-trait meta-analysis, a total of 29, 24, and 23 independent SNPs passed the threshold for significant association between BD and IBD, UC, and CD, respectively. We identified five novel pleiotropy genes including ZDHHC2, SCRN1, INPP4B, C1orf123, and BRD3 in both BD and IBD, as well as in its subtypes UC and CD. Pathway enrichment analyses revealed that those pleiotropy genes were mainly enriched in several immune-related signal transduction pathways and cerebral disease-related pathways. MR analyses provided no evidence for a causal relationship between BD and IBD.

CONCLUSION:

Our findings corroborated that shared genetic basis and common biological pathways may explain the comorbidity of BD and IBD. These findings further our understanding of shared genetic mechanisms underlying BD and IBD, and potentially provide points of intervention that may allow the development of new therapies for these co-occurrent disorders.
