Androgen receptor cofactors: A potential role in understanding prostate cancer

Prostate cancer (PCa) is witnessing a concerning rise in incidence annually, with the androgen receptor (AR) emerging as a pivotal contributor to its growth and progression. Mounting evidence underscores the AR's ability to recruit cofactors, influencing downstream gene transcription and thereby fueling the proliferation and metastasis of PCa cells. Although, clinical strategies involving AR antagonists provide some relief, managing castration resistant prostate cancer (CRPC) remains a formidable challenge. Thus, the need of the hour lies in unearthing new drugs or therapeutic targets to effectively combat PCa. This review encapsulates the pivotal roles played by coactivators and corepressors of AR, notably androgen receptor-associated protein (ARA) and steroid receptor Coactivators (SRC) in PCa. Our data unveils how these cofactors intricately modulate histone modifications, cell cycling, SUMOylation, and apoptosis through their interactions with AR. Among the array of cofactors scrutinised, such as ARA70β, ARA24, ARA160, ARA55, ARA54, PIAS1, PIAS3, SRC1, SRC2, SRC3, PCAF, p300/CBP, MED1, and CARM1, several exhibit upregulation in PCa. Conversely, other cofactors like ARA70α, PIASy, and NCoR/SMRT demonstrate downregulation. This duality underscores the complexity of AR cofactor dynamics in PCa. Based on our findings, we propose that manipulating cofactor regulation to modulate AR function holds promise as a novel therapeutic avenue against advanced PCa. This paradigm shift offers renewed hope in the quest for effective treatments in the face of CRPC's formidable challenges.

Tanshinone IIA destabilizes SLC7A11 by regulating PIAS4-mediated SUMOylation of SLC7A11 through KDM1A, and promotes ferroptosis in breast cancer

INTRODUCTIONBreast cancer (BC) is the most common malignancy in women with unfavorite prognosis.OBJECTIVESTanshinone IIA (Tan IIA) inhibits BC progression, however, the underlying mechanism remains largely undefined.METHODSThe cytotoxicity of Tan IIA was assessed by CCK-8 and LDH assays. Ferroptosis was monitored by the level of MDA, Fe²⁺, lipid ROS and GSH. IHC and western blot were employed to detect the localization and expression of SLC7A11, PIAS4, KDM1A and other key molecules. The SUMOylation of SLC7A11 was detected by Ni-beads pull-down assay and Co-IP. Luciferase and ChIP assays were employed to detect the direct association between KDM1A and PIAS4 promoter. The proliferative and metastatic properties of BC cells were assessed by colony formation, CCK-8 and Transwell assays, respectively. The in vitro findings were verified in xenograft and lung metastasis models.RESULTSTan IIA promoted ferroptosis by suppressing SLC7A11 in BC cells. Silencing of PIAS4 or KDM1A inhibited cell growth and metastasis in BC. Mechanistically, PIAS4 facilitated the SUMOylation of SLC7A11 via direct binding to SLC7A11, and KDM1A acted as a transcriptional activator of PIAS4. Functional studies further revealed that Tan IIA decreased KDM1A expression, thus suppressing PIAS4 expression transcriptionally. The inhibition of PIAS4-dependent SUMOylation of SLC7A11 further induced ferroptosis, thereby inhibiting proliferation and metastasis in BC.CONCLUSIONTan IIA promoted ferroptosis and inhibited tumor growth and metastasis via suppressing KDM1A/PIAS4/SLC7A11 axis.

Tanshinone IIA ameliorates cisplatin‐induced toxicology and cisplatin resistance via regulating SLC7A11 expression

AbstractCisplatin, a potent chemotherapy agent, is highly effective against various cancers but is hindered by resistance and toxicities. This study aims to investigate the roles of SLC7A11, a cystine/glutamate transporter, in cisplatin resistance, and explored Tanshinone IIA as a therapeutic option. Cisplatin reduced SLC7A11 in renal cells, worsening toxicity. Cisplatin‐resistant gastric cancer cells show increased SLC7A11, driving resistance, while SLC7A11 knockdown curbed resistance. Tanshinone IIA showed promise in alleviating cisplatin toxicity by enhancing SLC7A11 expression and reducing associated adverse effects, while it effectively reversed cisplatin resistance in gastric cancer cells by suppressing SLC7A11. Additionally, Tanshinone IIA counteracted cisplatin resistance by inhibiting PIAS4‐mediated SUMOylation of SLC7A11. Simultaneously, overexpressing miR‐375, which has been shown to target SLC7A11, exacerbated cisplatin toxicity via SLC7A11 downregulation, which Tanshinone IIA attenuates. In summary, our study unveils complex SLC7A11 regulation in cisplatin resistance and toxicity. Tanshinone IIA emerges as a promising modulator of SLC7A11 through individual pathways, offering novel insights into overcoming cisplatin resistance and reducing toxicities in cancer therapy.

合成致死WRN的变构抑制剂设计与发现

微卫星不稳定性（microsatellite instability，MSI）是指由于在DNA复制时插入或缺失突变引起的微卫星序列长度改变的现象，由错配修复功能（Mismatch repair, MMR）缺陷引起，是公认的重要致癌途径之一。MSI是一种遗传缺陷，常见于多种癌症中，包括结直肠癌、子宫内膜癌等。WRN（Werner syndrome RecQ helicase）作为一种RecQ家族的DNA解旋酶，在多项大规模基因组筛选证实是MSI癌症的合成致死靶点。 WRN参与合成致死的机制如下：当 DNA 在复制过程中发生错配，正常细胞主要由 DNA 错配修复蛋白（MSH2或 MLH1）进行 DNA 错配修复，而微卫星不稳定性肿瘤细胞由于缺乏错配修复基因（或错配修复系统存在缺陷），存在大量的TA重复序列，形成十字形的二级结构，使复制叉停滞，并引起ATR依赖的WRN磷酸化，来解旋二级结构及完成复制。当WRN缺失时，结构特异的MUS81-EME1核酸内切酶和SLX4一起切割二级结构以试图恢复复制叉的功能。但是，过多的MUS81切割最终导致了DNA末端切割，染色体破碎甚至细胞死亡。因此，发现WRN的抑制剂对于MSI肿瘤具有重要的治疗前景。为了获得该抑制剂，来自诺华公司的研究人员利用基于ATP结合的筛选方法，从15万个化合物中筛选到化合物1，并通过基于结构的药物设计，改善渗透性和脂溶性效率得到化合物；继而降低化合物logP并改善渗透性，引入邻甲基苯胺和羟基嘧啶得到化合物3和4。其中化合物4（HRO761）具有良好的物理化学性质和药代动力学，且无明显脱靶效应。共晶结构显示，HRO761作用于WRN解旋酶核心结构域，结合在D1-D2界面上的一个非保守变构位点，诱导D1和D2结构域发生约180°的相对旋转从而发挥功能。相互作用分析发现几乎所有HRO761上的重原子都参与了与WRN的极性相互作用，从而将WRN锁定在非活性状态。在高ATP条件下，HRO761在SW48细胞（MSI细胞）中测定的半数生长抑制浓度（GI50）为40nM，而对CAL33（microsatellite-stable, MSS细胞）无显著抑制作用。为了系统评估HRO761的细胞活性，他们对20个MSI（红色）和8个MSS（蓝色）细胞系中普筛了其抗增殖活性，发现HRO761在纳摩尔浓度(IC50:50-1000 nM)下选择性抑制MSI细胞系的克隆形成，且其敏感性与细胞内WRN的依赖性(DRIVE score)相关，对MSS细胞系无影响。在CellTiter-Glo（CTG）试验中, HRO761与WRN的结合在所有检测的MSI和MSS细胞系中都是相似的，其蛋白稳定化IC50(PS50)在10-100 nM范围内，而带有突变C727A或C727S会降低与HRO761的结合，并削弱其细胞增殖抑制和PD效应，说明尽管HRO761在所有细胞中都能有效结合WRN，但只在MSI细胞中抑制WRN从而产生选择性的细胞毒性。免疫印迹分析发现，HRO761处理MSI细胞后诱导产生DNA损伤反应，包括ATM、CHK2激酶磷酸化，p53激活和γH2AX增加等，而在MSS细胞系中未见此反应。同时还发现，HRO761可诱导WRN降解和染色质结合增加；蛋白质组学分析表明HRO761处理后的WRN蛋白丰度仅在MSI细胞系中显著下调。之前文献报道，当WRN受到抑制时，WRN在染色质上被困上并被PIAS4 泛素化修饰。泛素化的WRN被泛素E3连接酶RNF4识别，然后泛素化WRN导致p97/vcp介导的染色质提取并运送到蛋白酶体进行降解。RNA测序分析显示，HRO761在4个MSI细胞系中显著调控了多个基因的表达，包括CDKN1A、MDM2等p53靶基因，而在MSS细胞和WRN敲除细胞中没有观察到基因表达变化。因此，WRN抑制剂HRO761仅在MSI细胞中具有细胞毒作用，并诱导WRN蛋白降解。为了明确HRO761对MSI细胞的治疗作用，研究人员使用不同浓度HRO761 24h处理MSI细胞，发现其剂量依赖性激活ATM和CHK2、增加p21蛋白诱导并对p53依赖的基因呈剂量式调控。免疫荧光分析显示，随HRO761浓度升高，细胞核内绿色荧光显著增强，表明γ-H2AX含量明显升高，DNA损伤增加；细胞周期分析发现HRO761处理会使细胞阻滞在G2/M期。进一步机制研究发现，WRN降解并非细胞周期阻滞的原因，且经HRO761处理后，总CHK2蛋白水平虽有下调，但是由于在HRO761抑制生长的浓度并未导致CHK1/CHK2降解，说明HRO761致死不太可能是由于CHK2（或CHK1）丢失引起。体外实验显示，HRO761在HCT116 p53野生型和敲除型细胞中的IC50相似，且HRO761处理后两种细胞后均表现出抗增殖活性，表明HRO761作用机制不依赖于p53；同时，在p53野生型和敲除型细胞中，均可以观察到给药后ATM和CHK2磷酸化增加且WRN降解增加。上述结果说明，HRO761选择性诱导持续的DNA损伤反应，并引起细胞周期阻滞和凋亡，主要由ATM-CHK2通路驱动，而非依赖于p53。为了评价HRO761通过WRN的合成致死性能，研究人员对携带SW48移植瘤小鼠每日1次口服HRO761，发现给药后肿瘤增殖停滞，并在较高剂量下导致75-90%的肿瘤消退长达60天，之后肿瘤由于耐药性会出现剂量依赖性复发。在第7天及91天采集血样分析，HRO761的抗肿瘤功效与与血液中化合物浓度有关，且不会在体内蓄积，在GI90曲线上方的AUC与不同剂量下HRO761对肿瘤的疗效关系密切。小鼠给药后取样发现，HRO761在体内可有效抑制WRN，诱导DNA损伤反应（pKAP1增加）并激活p53通路(p21诱导)，这些变化在60 mg/kg剂量下给药后第8天后达到稳态，并随肿瘤消退而逐渐减弱。组织学分析也证实，HRO761剂量依赖性地抑制肿瘤细胞增殖（Ki67阳性），并引起细胞核增大。对不同MSI适应症的小鼠肿瘤模型和病人来源异种移植瘤（Patient-derived xenograft, PDX）模型进行大规模体内筛选，发现HRO761展现出广谱的抗肿瘤活性，其中2例完全缓解的模型分别为p53突变型（LS411N）和p53敲除型；同时。HRO761在p53敲除细胞中展现出比p53野生型细胞更好的抑制作用，再次印证了HRO761的p53非依赖性抗肿瘤机制。进一步，研究人员评估了HRO761与伊立替康联用的疗效。体外试验表明，HRO761与伊立替康在低于各自有效浓度时即可产生协同的细胞毒作用，并在高浓度HRO761处理下抑制了细胞再生长。体内研究证实，HRO761与伊立替康联用可在多个剂量组合下诱导肿瘤完全消退。这提示联合用药可能降低HRO761和伊立替康所需的个体给药剂量，从而拓宽治疗窗。综上，本研究运用化学筛选、药物设计等方法，得到具有高效抑制WRN活性的候选新药HRO761，评估了其在不同癌细胞系中的效果，验证了在MSI癌细胞模型中HRO761的合成致死效应，并对HRO761诱导合成致死效应的分子机制进行研究。目前HRO761正在开展I期临床试验，以评估其在结直肠癌等MSI实体瘤中的安全性和初步疗效。参考文献 1. van Wietmarschen, N., Sridharan, S., Nathan, W.J. et al.
