Eight metabolites were detected in the urine and feces of rats after oral and s.c. administration of DD 234 (methylscopolammonium methyl sulfate)(I) [18067-13-5].Characteristics of the metabolic pathway were that the quaternary ammonium moiety was unchanged, the major pathway involved oxidation at the para position of the benzene ring to introduce hydroxy and methoxy groups, and the minor products were the apo-derivative, glucuronide of I, and scopine derivativeAbsorption and excretion were examined by using 14C-labeled I, and the excreted total radioactivities were 82.2% of the oral dose and 93.6% of the s.c. dose in urine and feces.In respiratory air 14CO2 was not detectable.Comparison of the metabolic pathway with that of tertiary scopolamine was discussed.
