The study aims to provide representative phys. freeze-drying preformulation data on a series of oligonucleotides, i.e., phosphorothioate oligonucleotides.The compounds studied include GEM 91, GEM 92, GEM 132, and GEM 231, which were designed to treat various viral diseases and cancers through an antisense mechanism.DSC was performed with a unit having a "Modulated DSC" capability, and was used for freeze-dried powd. samples.The DSC thermograms on the freeze-dried solids were very complex, showing strong degradation exotherms, and enthalpy relaxation effects.The pure oligonucleotides and lactose formulations studied had moderate- to high-collapse temperatures and glass transition temperaturesThey freeze dry rapidly without collapse to produce solids of acceptable "elegance" and high class transition temperatures when dry.The results provided an excellent example of the power of Modulated DSC in the thermal anal. of complex systems.
